• Crypto – Crypto is the main protagonist in Destroy All Humans! 2 Reprobed and a member of an alien race known as the Furons. The voice actor who plays Crypto is Grant Albrecht.
• Natalya Ivanova – Natalya is Crypto’s love interest from Earth and a defector from the KGB. The voice actor who plays Natalya Ivanova is Courtenay Taylor.
• Orthopox-13 – Orthopox-13 is Crypto’s commanding officer who ends up dying in the sequel due to the mothership being destroyed by the KGB. The voice actor who plays Orthopox-13 is Richard Steven Horovitz.
• Arkvoodle – Arkvoodle is an ancient Furon God who came to Earth thousands of years ago and now holds a cult-like following on the planet. The voice actor who plays the “Lord of the Sacred Crotch” is Jim Ward.
• Shama Llama – Shama Llama is the head of the Cult of Arkvoodle who was rescued from the police by Crypto. He also possesses one of the strongest weapons in Destroy All Humans! 2 Reprobed. The voice actor who plays Shama Llama is Richard Tatum.

• The Freak – The Freak is a Bay City hippie who acts as an informant to the area. You’ll encounter him several times throughout the game and he’s rarely sober. The voice actor who plays The Freak is Richard Horvitz.
• Coyote Bongwater – Coyote Bongwater is a hippie commune leader dedicated to overthrowing “the man” and the creator of Revelade. He’s also one of the main antagonists in Destroy All Humans! 2 Reprobed. The voice actor who plays Coyote Bongwater is Richard Tatum.
• Prudence Kane – Prudence Kane is Coyote Bongwater’s girlfriend who firmly believes that Revelade will change the nation. The voice actor who plays Prudence Kane is Courtenay Taylor.
• Ivan Oranchov – Ivan Oranchov is the lead KGB agent in America and the one in charge of Bay City, who’s working towards destroying the country. The voice actor who plays Ivan Oranchov is Robin Atkin Downes.
• Dr. Go! – Dr. Go! is a Takoshimese scientist who knows a lot about the KGB and Black Ninjas in Destroy All Humans! 2 Reprobed. He’s also allied with the White Ninja clan and Crypto. The voice actor who plays Dr. Go! is Yuri Lowenthal.
• Sergei – Sergei is Natalya’s former lover, trainer, and Crypto’s archnemesis as a result. He too is curious about the KGB’s true motives. The voice actor who plays Sergei is Jim Ward.
• Gastro – Gastro is the main janitor and cook aboard the mothership. He also downloaded himself onto a Holopod to survive. The voice actor who plays Gastro is Phil Morris.

• The White Ninja Leader – The White Ninja Leader and his men worship the Furons and are considered to be the “good guys” of the ninjas. The voice actor who plays the leader of the White Ninja clan is Paul Nakauchi.
• The Black Ninja Leader – The Black Ninja Leader and his men ignited the war between them and the White Ninja clan. They’re also in with the KGB and viewed as the “bad guys” of the ninjas. The voice actor who plays the Black Ninja Leader is Steve Blum.
• Yamanasoku Hiroto – Yamanasoku Hiroto is the leader of the West Yakuza and the one who gives Crypto the quest to lure out Kenji Moto because of a comment made about his wife. Unlike the leader of the East Yakuza, Hiroto wears a peace sign on his suit.
• Kenji Moto – Kenji Moto is the leader of the East Yakuza who enraged Hiroto by claiming his wife is prettier.
